Integrin-mediated Protein Kinase A Activation at the Leading Edge of Migrating Cells Running Head: Integrins Specify Localized PKA Activation
نویسندگان
چکیده
cAMP-dependent protein kinase A (PKA) is important in processes requiring localized cell protrusion, such as cell migration and axonal path finding. Here we used a membrane-targeted PKA biosensor to reveal activation of PKA at the leading edge of migrating cells. Previous studies show that PKA activity promotes protrusion and efficient cell migration. In live migrating cells, membrane-associated PKA activity was highest at the leading edge, and required ligation of integrins such as α4β1 or α5β1 and an intact actin cytoskeleton. α4 integrins are Type I PKA-specific A-Kinase Anchoring Proteins (AKAPs) and we now find that Type I PKA is important for localization of α4β1 integrin-mediated PKA activation at the leading edge. متن کاملSpatial restriction of α4 integrin phosphorylation regulates lamellipodial stability and α4β1-dependent cell migration
Integrins coordinate spatial signaling events essential for cell polarity and directed migration. Such signals from alpha4 integrins regulate cell migration in development and in leukocyte trafficking. Here, we report that efficient alpha4-mediated migration requires spatial control of alpha4 phosphorylation by protein kinase A, and hence localized inhibition of binding of the signaling adaptor...
متن کاملThe non-receptor tyrosine kinase Lyn controls neutrophil adhesion by recruiting the CrkL-C3G complex and activating Rap1 at the leading edge.
Establishing new adhesions at the extended leading edges of motile cells is essential for stable polarity and persistent motility. Despite recent identification of signaling pathways that mediate polarity and chemotaxis in neutrophils, little is known about molecular mechanisms governing cell-extracellular-matrix (ECM) adhesion in these highly polarized and rapidly migrating cells. Here, we des...
متن کاملIntegrin-mediated activation of Cdc42 controls cell polarity in migrating astrocytes through PKCzeta.
We describe here a signal transduction pathway controlling the establishment of mammalian cell polarity. Scratching a confluent monolayer of primary rat astrocytes leads to polarization of cells at the leading edge. The microtubule organizing center, the microtubule cytoskeleton, and the Golgi reorganize to face the new free space, and directed cell protrusion and migration specifically occur p...
متن کاملRab11 is required for maintenance of cell shape via βPS integrin mediated cell adhesion in Drosophila
In eukaryotes, vesicle trafficking is regulated by the small monomeric GTPases of the Rab protein family. Rab11, (a subfamily of the Ypt/Rab gene family) an evolutionarily conserved, ubiquitously expressed subfamily of small monomeric Rab GTPases, has been implicated in regulating vesicular trafficking through the recycling of endosomal compartment. In an earlier communication, we have shown th...
